The 144,000 are of God's annointed (or chosen) to rule with Jesus in Heaven during the thousand year reign, after the wicked are destroyed on earth. During this time mankind will grow to perfection, the Earth will be made a paradise.
I'd talk more but that gets into explaining a pretty lengthy and I don't want to come off as preachy to anyone.
